摘要:

目的观察大蒜素对气道平滑肌细胞(ASMC)。一平滑肌肌动蛋白(a-S M A)表达的影响,以评价大蒜素对气道重塑的抑制作用,并探讨其相应的作用机制。方法用。,2.5,5.0,10.0 p,mol/L的大蒜素处理人ASMC细胞48 b后提取细胞总RNA和蛋自。荧光定量聚合酶链反应(PC R)检测细胞。-SM A mRNA表达,蛋白质免疫印迹法(Western blotting)检测细胞。-SM A和磷酸化Smadl ( p-Small 1)的蛋白表达。结果随着大蒜素处理ASMC细胞浓度的提高(0,2.5,5.0,10.0林mol/T.),细胞。-SVLA mRNA表达呈进行性下降(分别为0.543士0.121,0.354士0.072 , 0.223士0.058,0.191士0.034,各剂量间两两比较差异有统计学意义(均尸< 0.05 ) ; a-SM A和p-Small 1的蛋白表达也呈进行性下降〔a-SMA蛋白(灰度值比值)分别为。.96士0.02 , 0.72士。.16, 0.54士0.11,031士0.14 , p-Smadl蛋自(灰度值比值)分别为0.94士0.03 ,0.76士0.13 , 0.62士0.11 ,0.43士0.12,各剂量间两两比较差异有统计学意义(均P< 0.05 )。结论大蒜素可以剂量依赖性地下调AS}TC细胞a-SMA的mRN.A和蛋白表达,抑制,nad蛋自的磷酸化,阻止转化生民因子书(TGF-团和Rho激酶信号通路。   To insestigate the effectf)fallicin on the expression of a一*month muscle actin(-SMA) in airway smooth muscle cells (ASMC),and to evaluate the mechanism of allicin。,:inhibition of airwayremodeling.
